The South Carolina State Ports Authority (SCSPA) oversees Port Royal and the implementation of the Port Royal Tract Development Agreement between the Town of Port Royal, the State of South Carolina, and the SCSPA. The agreement makes redevelopment of the port a top priority. The 2004 "A New Vision for the Port" study outlined how Port Royal would preserve and expand the town's traditional character into the port by promoting mixed land use and residential types. The Agreement covers an area of about 128 hectares that includes 20 hectares of highlands.

The Port Royal Landing Marina is located on the Intracoastal Waterway. Open all year, it offers almost 183 meters (600 feet) of dock face with easy access to the Atlantic Ocean through both Port Royal and St. Helena Sounds. From April 1 through November 30, both the marina office and fuel dock are open every day from 8am until 6pm. Off season, they are open until 5pm daily. Annual and transient boaters at the Port Royal Landing Marina have free access to indoor and outdoor pools; a spa with sauna and steam room; a fitness center with fitness classes; a gymnasium; and health and wellness programs. The marina offers a courtesy car to take boaters to local shops during office hours.

The Port Royal Landing Marina has floating concrete docks with electric power and freshwater hookups at all slips. The marina can accommodate boats to 36.6 meters (120 feet) with 4.6 meters (15 feet) depth at MLW. Valvtect fuel and gas are available at the Port Royal Landing Marina. There are vacuum pump-outs at the fuel docks, and portable electric pump-outs are available.

Other amenities at the Port Royal Landing Marina are code-locked showers and restrooms, a 24-hour laundry facility, and Wi-Fi Internet service. The Back Porch Grille is located on-site, offering a 10% discount for transient boaters. The ship's store offers free overnight delivery. The Port Royal Landing Marina has a covered deck, a grill area, and an open green space. Marina customers also have access to in-shore/and off-shore fishing charters, car rentals, taxicabs, and River Safari Nature Tours.
